Abstract | Different types of explant (whole leaves, leaf segments, petioles and internodal stem pieces) excised from in vitro growing seedlings of white guinea yam (Dioscorea rotundata Poir.) cv. Obiaoturugo, were cultured on MS medium supplemented with increasing concentrations of NAA. At lower concentrations (0.5 to 1.0 mg/l) NAA induced plantlet regeneration from petiole and internodal stem segments at a frequency of 40% of cultured explants, and rooting with no or little callusing on whole or segmented leaves. At higher NAA concentrations, all types of explant reacted towards callus formation. The induced calli developed plantlets when subcultured in MS medium supplemented with 2 mg/l BAP and 0.1 mg/l NAA. Direct regeneration from petioles and stem pieces and indirect regeneration though callus induction, are both regarded as possible tools for yam improvement. |
